Cell Culture Policy

The cell culture area of the SCIF is open to investigators utilizing SCIF's resources. Usage of the biological safety cabinets, incubators, and associated equipment is currently free of charge. The available equipment includes:

  • Nuaire Biosafety Cabinets
  • Thermo Cell Culture Incubators - 5% CO2, humidified
  • Nikon inverted fluorescent tissue culture microscope - green (GFP) and red (Tomato, Cherry, etc) filters
  • Sorvall hanging bucket centrifuge - adaptors for plates, 50ml, 15ml, 5ml and 1.5ml tubes
  • 37C water bath
  • diH2O system
  • 4C refrigerator
  • -20C freezer

All users are required to bring their own consumable products, including tips, pipettes, tubes, media, and dishes.

  • Space will be assigned to store consumables. 

Users will be responsible for leaving the culture area clean. Please perform these tasks to ensure the space stays orderly:

  • Empty tip waste in the hood, spray jar with 70% EtOH and wipe out
  • Spray down the hood with 70% EtOH and wipe down, wipe the glass
  • Clean vacuum line with 70% EtOH – don’t leave media in the line
  • Fill EtOH spray bottles
    • Use 70% histoprep Ethanol under the sink
  • Check waste flask to make sure it’s not past the fill line
    • To dump: Add bleach (under sink) to 10%, wait 20 minutes and dump down sink. It should go from pink to clear.
    • Place ~500ml of bleach in the bottom of the empty flask and put back
  • Turn off centrifuge
  • Turn off microscope
  • Clean your stuff off counter tops
  • Check incubator and water bath water level – notify SCIF if needing refill
  • Check biohazard waste – notify SCIF if full
  • Check CO2 tanks – notify SCIF if running low
  • Place regular trash in the hallway if it’s full – custodians only dump if it’s in the hall.
